All, question:
Can you successfully use the HR10-250 and Tivo services with regular DirecTV\Tivo service & OTA HD, not subscribed to the DirecTV HD service? If so, will it actually tune the regular DirecTV channels, OTA HD channels, and update the guide accordingly? Will it dual tune both the DirecTV Sat signals and OTA HD signals?

Assuming you have your locals, and therefore get programming information for those channels from the satellite, the answer to all of your questions is yes. If you don't, I believe you could still record OTA, but would have to do so manually w/o the name of the programs appearing in your "Now Playing" list.
I am currently doing precisely what you're asking, so I know it'll work.

Yes. That is how I use it!
Just to clarify, it is a dual tuner. It can tune to two different incoming signals at one time, any combination of the four inputs (2 Sat and 2 OTA).

You don't need to subscribe to DirecTv locals to record OTA. The guide data will still show your OTA info.

You'll be fine, there's no need to purchase the HD package from D* unless you want the progamming. The HR10 will be fine with SD programming from D* plus your OTA locals. You'll get the guide data for your OTA channels. You can simultaneously record 2 OTA or 2 sat or one of each.
